i am finally releasing the video on
mk677
so i've spent many hours looking at the
research around
mk677 so that i could put it together in
a summarized format for you all to
inform you about everything that there
is or almost everything that there is so
essentially this video is everything you
need to know about mk677
or almost everything you need to know
because i'm sure i've missed out on a
few things
so the title of the video is just an
mk677 only cycle
that's more to draw in attention but i
will briefly touch on that
but for the most part i'm just going
over everything about mk 677 so that you
guys know what you're taking
so mk 677 is known as a growth hormone
secretary however it doesn't really work
via that pathway it's more of a ghrelin
receptor agonist and it increases the
level of ghrelin which secretes growth
hormone
but essentially all you need to know is
that it does increase the levels of
growth hormone and why is growth hormone
so beneficial well it causes lipolysis
or breakdown of fat as well as
increasing muscle mass and a lot of big
bodybuilders like this drug because it
keeps them lean while putting on size
so there are many growth hormone
receptors but the main actions i don't
want to say main actions but a lot of
the effects that we see in bodybuilders
come mainly from igf-1 now igf-1 is
stimulated by growth hormone and we
measure igf-1 to see if the growth
hormone is working so growth hormone
essentially stimulates these receptors
on the liver and this creates igf-1
so growth hormones half-life is quite
short and it's secreted in like a
pulsatile manner because of this short
half-life however igf-1 stays in the
bloodstream a lot longer and when
looking at these drugs it's important
for us to look at how long the igf-1
levels are elevated not growth hormones
since igf-1 is more important if from a
bodybuilding aspect why were things like
mk677 created
well we have been using growth hormone
for a long time especially in children
with short stature however there were
concerns about the safety around growth
hormone especially with regards to the
increase in mortality and older studies
that they found however nowadays they
found that that may not be the case but
there are a lot of side effects like
bone cancer prominence was increasing so
they wanted to see if they could make
something a bit more selective
so essentially they started making these
growth hormone releasing peptides that
was initially made and the issue with
them is whilst they were a tad bit more
selective
they required frequent injections and
another issue was that it was poorly
orally bioavailable
but over time and over a lot of
experimentation they eventually found
this substance that could stimulate the
growth hormone releasing peptide 6
receptor
and this was a derivative of a
a benzolactin
and it had a very long name but
essentially this came to be what we know
as mk677
and the favorable thing about mk677 is
it had very high oral bioavailability so
they started off safety trials in
beagles and unfortunately a lot of our
studies on the pharmacokinetics are
based on beagles and we haven't really
done many in humans but essentially in
this two-week trial they found it to be
safe and then it proceeded to go into
further development so i always like
touching on the pharmacokinetics so you
can understand proper dosing protocols
and it is stated but remember this is in
beagles that mk 677 has a half-life of
about four to six hours so that's mk 677
itself but in these studies they noted
that the important factor which is igf-1
is seems to be elevated for about 24
hours
so what this means is that if you want
that igf-1 level to be constantly
increased you need to do it every day or
dose it every day they found that if you
did every other day dosing in these
beagles that you would not get the
significant increase in igf-1 levels and
this increase was about
124 percent or 26 sorry so just remember
there was no increase noted on alternate
day dosing which means it has to be
dosed every day so let's look at the
safety of this drug and obviously you
want to see if it's safe in children
because that's why it was kind of
designed
obviously there are other purposes of it
too for
elderly individuals and people like that
so in one of the first studies on
mk677 in children they used a dose or
they dusted up to 0.8 milligrams per kg
a day and they found that this rain this
dose was effective to significantly
raise growth hormone as well as igf-1
and it did not interfere with any of the
other hormones like prolactin or messing
around with the thyroid hormones however
in the study they did not look at growth
specifically
i'm not going to look at growth either
because this is for adults not this
video is for adults not children
so let's see if mk 677 is anabolic as
everyone says it is
so in this one study in 1998 they looked
at individuals in a catabolic state so
in a state where they were essentially
in a deficit and muscle protein
synthesis was or muscle protein
breakdown was greater than that of
synthesis
so in this trial they were given 25
milligrams they were compared to placebo
and what they found was that in this
trial
the individuals who were taking mk 677
had a positive daily nitrogen balance
even though they were fasting so the
individuals were forced to fast during
this period to put them in a catabolic
state
and it found that the mk-677 group
actually had a positive nitrogen balance
which means that they had a positive
protein balance or muscle balance
not
not really but that's essentially what
it means and in the placebo group they
actually had quite a significant
negative nitrogen balance
so this essentially demonstrated that
mk677
preserve muscle mass or tissue in
catabolic states which is a good sign
for the elderly and people with wasting
conditions furthermore not much
important to any of you guys since um i
doubt any of you have osteoporosis or
osteopenia but it did show that it was
very favorable in bone positive bone
remodeling so good for the elderly
especially if they had fractures
and when they looked at individuals with
fractures it did improve the markers of
osteoporosis however functional outcomes
were not that greatly improved
what was interesting though was in the
study igf-1 levels were significantly
increased in comparison to placebo and
this again was in 20 using a dose of 25
milligrams
so in the
in the mk-677 group of 25 milligrams a
day
the increase in igf-1 levels were 84
percent and in placebo 17
and the 84 did reach statistical
significance
but the two studies i just mentioned
were in elderly individuals what about
in healthy men
so there aren't many studies on this but
there was a double blind randomized
control trial which we do like because
we get to see if there's causation as
opposed to correlation
and in this study they had 24 men and
they found that in the mk 677 group when
compared to placebo they gained three
kilograms of fat-free mass more than the
placebo group
and this was again at 25 milligrams a
day however they did not show any change
in abdominal or visceral fat mass which
you'll see often reported since growth
hormone is actually
has actually been shown to decrease
visceral and abdominal fat mass
of note and what many people will
mention is that in the study the serum
prolactin so prolactin levels were
greatly or significantly increased just
like the growth hormone levels
but we'll touch on that again later
and we in fact do have long-term trials
which is good from a safety point of
view so there was a double blind
randomized control trial which is good
on 65 healthy elderly individuals and
they lasted two years and they found
that in the group using 25 milligrams of
mk-677 a day growth hormone as well as
igf levels were equal to that of their
healthy of healthy individuals and
you'll see a lot of people reference the
study
of note also is that fat free mass
increased in the mk 677 group they all
gained
on average a kilogram of lean muscle
mass
in contrast to the placebo group which
actually lost fat free mass
of interest also is that their ldl
cholesterol the bad cholesterol also
decreased
however this fat free mass increase was
not accompanied by any increase in
strength
or fat loss now a side effect or
positive that many individuals will note
when taking mk 677 and that is sleep and
this has been demonstrated in trials in
a smaller crossover study in eight young
males they found that
in this study those taking 25 milligrams
of mk67 the seven a day had a 50
increase in stage four sleep duration
and if i'm not mistaken stage four sleep
is essentially
deep sleep the sleep before rem sleep
and these individuals also had a 20
increase in rem sleep but what was
interesting is that in older individuals
there was a massive increase in the
benefit that it had on sleep and that
was demonstrated by elderly individuals
having a 50
increase in rem cycle sleep but let's
look at the side effects
so
many may look at these studies and see
that mk 677 in the one study increased
the risk of congestive heart failure or
heart failure however not many people
understand what heart failure is firstly
these individuals in the group were
already
elderly not young and healthy
furthermore they had high baseline blood
pressure
and lastly heart failure is not really
an acute condition
people commonly think heart failure and
heart attacks are synonymous but
essentially heart failure is more of a
disease
of chronic conditions such as
chronically high blood pressure
previous
heart attacks can lead to heart failure
which is where the heart just doesn't
have enough strength to pump a heart
attack is where muscle in that
in the myocardium dies
and what happens is when you get older
the increase you have an increased risk
of getting a weaker heart and something
like
mk677 which will increase your water
retention as well as possibly your blood
pressure can precipitate
can precipitate
decompensation of an already weakened
heart
however in the study
they noted that the prevalence was about
6
in these elderly individuals and if you
look at the prevalence of heart failure
in elderly individuals as a whole
it's six percent
so i think it is important to note it
however although you won't get heart
failure unless it's chronic like over
the course of many years
you won't get heart failure using mk677
but the side effects can precipitate
heart failure or
or lead you to having an enlarged heart
over the course of
a few years how it does this we're not
too sure it's probably similar to that
of growth hormone by increasing the ras
system water retention increasing your
blood pressure things like that can lead
to a weakened or enlarged heart which
leads to
heart failure
however in many other studies and
healthy elderly individuals one with 161
elderly individuals over six months
noted no adverse effects
as well as in another smaller trial with
32 healthy in elderly individuals they
also noted no adverse effects
however in the two-year trial that we
mentioned previously one of the effects
that was noted quite commonly or 67
percent of the individuals actually
noted this is that they had an increased
appetite and this isn't surprising since
it stimulates ghrelin the hunger hormone
of interest there were fewer deaths in
the mk-677 group but when confounding
variables were controlled for it didn't
appear to seem to be related to the
mk677
now one side effect that is demonstrated
in all studies and i would recommend or
i think is important to know and that is
what it does to your blood glucose or
sugar
so it causes elevated sugar levels
and it does this through decreasing your
insulin sensitivity
so it's quite commonly demonstrated that
in these studies that the individuals
taking mk-677 had higher fasting glucose
levels
and this led to an increase in hba1c
levels so hba1c is your glycated
hemoglobin and essentially is a marker
for
long-term glucose control
and that was increased in the
individuals taking mk-677
it did reach statistical significance
and this was at a dose of 25 milligrams
so i think it is important to note
whilst i don't think
it would have much of an impact on a
healthy younger individual if you are
diabetic or our elderly i would take
this into account and take precautionary
measures whether it be through dietary
manipulation the use of anti-glycemic
agents such as metformin i think that's
important to take into consideration
other side effects include increased
prolactin levels which we mentioned
it is a significant increase but whether
or not it will be significant in you and
you will become symptomatic is really
individual dependent i would recommend
getting blood tests if you do think you
have symptoms of high prolactin levels
a big issue is water retention so i
would limit your salt intake and ensure
your blood pressure is controlled as
this was probably the main reason for
those cases of heart failure in those
studies people have mentioned that free
testosterone decreases which has been
shown in a study and it did reach a
level of significance however it did not
make the participants hypogonadal the
decrease just reached statistical
significance it didn't decrease it to
the point of becoming hypogonadal
does it cause growth hormone suppression
well actually these secretogs were
created in order not to inhibit the
pituitary and hypothalamus they they
work at the pituitary and hypothalamus
but don't cause negative feedback
so after stopping the secretogs there is
no need to do a growth hormone pct
as they do not cause negative feedback
and do not cause your intrinsic growth
hormone production levels to decrease do
you need a pct for your hormones such as
testosterone no since it does not
significantly impact
those hormones some recommend a pct with
metformin that is really up to you and
whether or not you feel your fasting
glucose levels were impacted greatly but
again you'd have to get that checked you
can't go off feeling
so that is almost all there is on mk677
so far i tried my best to get all the
data on humans as i don't like looking
at data in rats and animals
as a lot of the time it's not really
applicable to humans but it's a it
starts off as a good basis but once we
have human trials we'll use those
instead
so if you are looking to run this as a
mk677 only cycle the dosages they seem
to use are 25 milligrams again i'm a
doctor so i don't recommend this but 25
milligrams seems to be the doses that
was used with minimal adverse effects
the length of your cycle is really up to
you as we have demonstrated that
up for up to two years it was used
safely with not many adverse effects and
it did have an increase in fat-free mass
but the increase in strength and
decrease in abdominal fat wasn't really
demonstrated in these studies
but just keep an eye on your blood
pressure as water retention is quite
commonly reported so let me know what
